The pHT43 vector is a high-level expression vector for recombinant protein production and secretion into the culture medium. The expression is based on the strong σA-dependent promoter preceding the groE operon of B. subtilis which has been converted into an efficiently controllable (IPTG-inducible) promoter by addition of the lac operator. The amyQ signal sequence (encoding for the signal peptide of a-amylase) is located downstream of the Shine-Dalgarno Sequence, followed by restriction sites for cloning the gene of interest. pHT43 is an E. coli / B. subtilis shuttle vector, that provides ampicillin resistance to E.coli and chloramphenicol resistance to B. subtilis.
